Description
PNC-27 is an experimental peptide that joins a short segment of the tumour-suppressor protein p53 (residues 12–26) to a membrane-residency sequence. It was designed at SUNY Downstate to study how cancer cell lines that carry the protein HDM-2 in their outer membrane respond to a p53-derived probe. Pure Peptides Canada supplies it only as a lyophilized laboratory reagent in 30 mg vials. It is not a cancer treatment, not an approved medicine and not for human use.
PNC-27 at a glance
- What it is: p53(12–26), the region that binds HDM-2 (MDM2), linked at its C-terminus to a penetratin-type membrane-residency peptide.
- What researchers study: HDM-2 in cancer-cell membranes, peptide-induced membrane pore formation, and selectivity between tumour and non-tumour cells in culture.
- Evidence level: cell-line and mouse studies, almost entirely from the group that developed it. No published controlled human trials.
- Supplied as: sealed lyophilized powder, 30 mg, single vial or box of 10.
- Delivery: ships from inside Canada by tracked Xpresspost in 3–7 days ($25 CAD), or free in 4–12 days on orders over $299.
Regulatory status: PNC-27 is an experimental compound that has not been approved anywhere. It is not a treatment for cancer or any other disease. We sell it strictly as a laboratory reagent for in-vitro research to qualified research buyers, and we do not provide dosing, administration or medical information.

What is PNC-27?
Inside cells, the protein HDM-2 (MDM2 in mice) binds the N-terminal region of p53 and marks it for destruction. Matthew Pincus, Josef Michl and colleagues at SUNY Downstate took the p53 segment that contacts HDM-2, residues 12 to 26, and attached a membrane-residency sequence so the peptide would sit in cell membranes. They report that many cancer cell lines, unlike the normal cells they tested, carry HDM-2 in their outer membrane, and that PNC-27 binds it there and forms pores that kill the cells by necrosis rather than apoptosis.
In a 2014 study in Annals of Clinical and Laboratory Science, the group reported that PNC-27 killed K562 leukemia cells, which lack p53 entirely, while leaving control leukocytes unaffected, suggesting the effect does not depend on the cell’s own p53. The same group has reported mouse xenograft results. These findings have not been confirmed in controlled human trials, so PNC-27 remains a research tool for studying membrane HDM-2 and peptide–membrane interactions.
What PNC-27 research looks at
- Membrane HDM-2: where and in which cell lines HDM-2 appears at the cell surface.
- Pore formation: membranolysis, LDH release and necrosis versus apoptosis markers.
- Selectivity: tumour versus non-tumour cell responses, with PNC-29 as a negative control.
- p53-pathway peptides: comparisons with other p53-interacting probes such as FOXO4-DRI, and with membrane-active peptides such as LL-37.

Where can I buy PNC-27 in Canada?
Pure Peptides Canada supplies PNC-27 only as a lyophilized laboratory reagent in 30 mg vials, shipped from inside Canada by tracked Xpresspost in 3–7 days ($25 CAD), or free in 4–12 days on orders over $299. It is not sold for human use.
Is PNC-27 a cancer treatment?
No. PNC-27 has been studied only in cell lines and animal models, it has not been approved anywhere, and there are no published controlled human trials. We supply it strictly for in-vitro research and provide no medical information.
What is the difference between PNC-27 and PNC-28?
Both join a p53 segment to the same kind of membrane-residency sequence. PNC-27 uses p53 residues 12–26; PNC-28 uses the shorter segment 17–26.
How do I confirm the identity of a PNC-27 batch?
Published descriptions of the membrane-residency segment vary, so ask us for the full sequence and expected mass for your lot, then check that the mass-spectrometry result matches it. Our COA guide explains what else to look for.
Can PNC-27 be used in people or animals?
No. This product is for in-vitro laboratory research only and is not a drug, supplement or food. We do not provide dosing or administration information.
For laboratory research use only. Not a drug, supplement, food or cosmetic, not for human or veterinary use, and not intended to diagnose, treat, cure or prevent any disease.

John M. –
battling cancer, tired of chemo and side effects of it, but still playing russian roulette with sepsis on this PNC-27 since its grey market. At this point, im open to whatever and this specifically targets the cancer cells and not the healthy ones… wish me luck! no complaints about it so far and it seems promising.
