Description

FOXO4-DRI is a cell-penetrating peptide built from D-amino acids in reverse order, designed to break the bond between FOXO4 and p53 inside senescent cells. It became one of the best-known senolytic research tools after a 2017 Cell paper. Pure Peptides Canada supplies it as a lyophilized research reagent in 10 mg vials, with an independent Janoshik HPLC and mass-spectrometry report, CAD pricing and tracked shipping from inside Canada.

FOXO4-DRI at a glance

  • What it is: a D-retro-inverso peptide that mimics part of FOXO4 and competes with it for binding to p53, linked to a cell-penetrating sequence.
  • What researchers study: selective apoptosis of senescent cells, the FOXO4–p53 axis, chemotherapy-induced senescence and tissue function in aged mice.
  • Evidence level: cell and mouse studies, centred on the 2017 study. No published controlled human trials.
  • Supplied as: sealed lyophilized powder, 10 mg, single vial or box of 10.
  • Delivery: ships from inside Canada by tracked Xpresspost in 3–7 days ($25 CAD), or free in 4–12 days on orders over $299.

Where the evidence stands

  1. Cell studies
  2. Animal models
  3. Controlled human trials
  4. Approved medicine

Evidence rests largely on one landmark mouse study and follow-up cell work. FOXO4-DRI is not an approved medicine.

What is FOXO4-DRI?

Senescent cells stop dividing but resist dying, and their accumulation is linked to tissue ageing. Peter de Keizer’s group at Erasmus MC in Rotterdam found that in senescent cells the transcription factor FOXO4 holds p53 in the nucleus in a way that keeps the cell alive. They designed a peptide to interrupt that interaction. Building it from D-amino acids in reverse order, the D-retro-inverso approach, keeps the side chains in a similar orientation while making the peptide largely immune to protein-digesting enzymes.

In the 2017 Cell study, FOXO4-DRI pushed senescent cells into apoptosis while sparing normal cells, and in mice it counteracted chemotherapy-induced toxicity and improved fitness, fur density and kidney function in fast-ageing and naturally aged animals. Those results drew wide attention, but they have not yet been confirmed in published human trials, and questions about mechanism and selectivity remain open.

What FOXO4-DRI research looks at

  • Senolysis: selective apoptosis of senescent versus proliferating cells.
  • FOXO4–p53 signalling: p53 nuclear exclusion and downstream apoptosis.
  • Chemotherapy-induced senescence: doxorubicin and related models.
  • Ageing models: tissue function in fast-ageing and naturally aged mice.
  • Peptide design: D-retro-inverso stability and cell-penetrating delivery.

Regulatory status: FOXO4-DRI is not an authorized drug in Canada. We sell it strictly as a laboratory reagent for in-vitro research.

FOXO4-DRI FAQ

Where can I buy FOXO4-DRI in Canada?

Pure Peptides Canada stocks FOXO4-DRI as a lyophilized research reagent in 10 mg vials and ships it from inside Canada by tracked Xpresspost in 3–7 days ($25 CAD), or free in 4–12 days on orders over $299.

What does DRI mean?

D-retro-inverso: the peptide is made of D-amino acids arranged in reverse order. This keeps a similar side-chain layout to the natural sequence while making the peptide highly resistant to enzymatic breakdown.

What is a senolytic?

A compound that selectively kills senescent cells, the damaged cells that stop dividing but resist normal cell death. FOXO4-DRI is one of the best-known peptide senolytics in research.

Is this FOXO4-DRI third-party tested?

Yes. An HPLC purity and mass-spectrometry identity report from Janoshik Analytical, an independent lab, is listed on our Lab Testing page. Our COA guide explains how to read it.

Can FOXO4-DRI be used in people or animals?

No. This product is for in-vitro laboratory research only and is not a drug, supplement or food. We do not provide dosing or administration information.

For laboratory research use only. Not a drug, supplement, food or cosmetic, not for human or veterinary use, and not intended to diagnose, treat, cure or prevent any disease.

Storage & Handling

Store sealed lyophilized vials cold and away from light to preserve stability. Keep vials sealed until use and follow any batch-specific notes on the Certificate of Analysis. Cold-chain shipping is used where a product requires it.

Because all products are sold strictly for laboratory research use, we do not provide reconstitution, dosing, or administration guidance of any kind.

Certificate of Analysis

A Certificate of Analysis (COA) is published for the batch you receive, reporting purity (HPLC) and identity (MS). Every batch is tested by Janoshik Analytical, an independent third-party lab. Match the batch number printed on your vial to its certificate on our Lab Testing page.
